Cleaning and Operation Procedure for Centrifuge Machine |
1.0 OBJECTIVE To lay down a procedure for cleaning and operation of centrifuge machine. 2.0 SCOPE This SOP is applicable for cleaning and operation of centrifuge machine in Quality Control Department. 3.0 RESPONSIBILITY QC-chemist/QC officer/ Sr. officer / Executive / Sr. Executive 4.0 ACCOUNTABILITY Head-QC 5.0 PROCEDURE 5.1 Definition 5.1.1 A centrifuge is a mechanical or electromechanical device to separate various components of a liquid. It achieves that by simulating a very high gravity environmental inside the tubes by producing centrifugal forces by spinning very fast in quality control department. 5.2 Operation: 5.2.1 Control panel 5.2.1.1 Above image show the display board of the centrifuge model No. R-8C plus. It consists of two displays, 2 LEDs and 5 keys. 5.2.1.2 RPM display: it is 4digit display which shows the actual RPM in run mode (centrifugation) and the set RPM while normal mode. 5.2.1.3 Timer display: it is 2-digit display showing the set time in normal mode and the time down count in run mode. 5.2.1.4 LEDs: • Motor LED: this is a dual LED which is RED while in normal mode and it turns green when the motor (START/STOP). • DC break LED: This is a dual LED which glows RED when DC break function is used, otherwise it remains green. 5.2.1.5 Key functions 5.2.1.5.1 Start/Stop: • In normal mode pressing the key takes the device into “centrifugation mode” • In “centrifugation mode” key is used for stopping the process and coming back to normal mode. 5.2.1.5.2 Set key • Press the key in “Normal Mode” takes the device into “Set-up mode” and the key used to set the values of parameters in “Set-up mode” 5.2.1.5.3 Break key • Key is used to enable/disable the DC break feature selection in “Normal” or “centrifugation” mode. 5.2.1.5.4 Increment key • Key is used to increase the value of parameter to be set while in “Set-up” mode. 5.2.1.5 Decrement key: – key is used to decrease the value of parameter to be set while I “set up” mode. 5.2.1.6 DC brese. 5.2.2 Operating instruction 5.2.2.1 Centrifuge machine have four modes 1) operation modes 2) normal mode 3) set-up mode 4) centrifugation mode. 5.2.2.2 List of parameter • RPM: sets the RPM at which the centrifugation is to carried out settable between 50 to 6000 in steps of 10. • Time: sets the time for which the process is to be carried out settable between 0 to 99 minutes. 5.3 Operation method 5.3.1.1 Ensure that the instrument is clean and free from dust. 5.3.1.2 Switch “ON” the mains and instrument power button. 5.3.1.3 Open the upper li by releasing the lock and lifting it up. 5.3.1.4 Place the centrifuge tubes in the compartment provided for it. 5.3.1.5 The compartment is designed in such a way to place four centrifuge tubes at a time. 5.3.1.6 Adjust the RPM speed by the button provide on the front panel of the instrument. 5.3.1.7 A scale with a deflection display is provided on the front side panel of the instrument. This scale can measure the RPM form to 0 to 6000. 5.3.1.8 Set the required time by the timer setting button having 0 to 99. 5.3.1.9 Switch “OFF” the mains after usages. 5.4 Safety instruction 5.4.1 Only trained personnel are allowed to operate this centrifuge 5.4.1.1 Proper handling of the instrument. 5.4.1.2 Ensure level and stability. 5.4.1.3 Balance centrifuge tubes equally. 5.4.1.4 Ensure use of rubber cushions for glass tubes. 5.4.1.5 Bring speed knob to ‘OFF’ and increase speed gradually. 5.4.1.6 Check carbon brushes periodically. 5.5 Cleaning of instrument 5.5.1 Clean rotor, buckets and accessories outside of the centrifuge bowl. 5.5.2 Separate all rotors, buckets, lids, adapters and tubes to allow through cleaning. 5.5.3 Rinse rotor and all accessories with warm water and a detergent that is suitable for use with the materials. If in with the material 5.5.4 Use a soft brush without metal bristles to remove stubborn residue. 5.5.5 Rinse rotor and all accessories with distilled water 5.5.6 Dry all of the rotor and accessories after cleaning with a cloth or in a warm air cabinet at a maximum temperature of 50°c. |
