1.0 OBJECTIVE To lay down a Procedure for cleaning, operation and qualification of Bio-safety cabinet. 2.0 SCOPE This procedure is applicable for cleaning, operation and qualification of bio-safety in the Quality Control Department of Microbiology Area. 3.0 RESPONSIBILITY • Microbiologist: is responsible for carrying out the established procedure • Head of Microbiology : is responsible for ensuring that the SOP is followed correctly. And ensuring that biosafety cabinet are validate before due date. 4.0 ACCOUNTABILITY • Department Head • Head Quality / QA To Read this SOP click here SOP for Growth Promotion Test and Inhibition Test of Media. 5.0 PROCEDURE 5.1 Cleaning procedure 5.1.1 Use lint free cloth dipped in filtered 70% IPA solution so as sanitize all surface of the biosafety cabinet in a straight and superposed way. The bench top from the back to front and the sides, from upwards to down (frequency: before and after working). 5.2 Starting of bio-safety cabinet 5.2.1 Switch ON the main supply form the main circuit placed on the front panel of the instrument. 5.2.2 There are three switch and two pressure manometer and one UV burning meter in the front panel of instrument 5.2.3 Switch (1) UV light (2) tube light (3) blower. 5.2.4 Check the pressure manometer level should be zero when blower is in “OFF” condition. 5.2.5 Press the blower switch “ON” the blower sterile the inner surface with 70% IPA solution, switch “OFF” blower and finally switch “ON” the UV light for not less than 30min. 5.2.6 After 30min. switch “OFF” the UV light and switch “ON” the tube light and blower for 5-10 minutes before carry out the analysis under bio-safety cabinet. 5.2.7 Check the pressure manometer, the level of manometer should be between in 10-20 when the blower is “ON” condition. 5.2.8 Also check Exhaust manometer reading should be between in 5-8mm when the bio-safety is “ON” condition. 5.2.9 Confirm the inward airflow by holding a tissue at the middle of the edge of the viewing panel and checking if it gets pulled in. 5.2.10 Assemble material introduce only those items that are required to perform the procedures and arrange in a logical order. Each item should be wiped with a disinfectant prior to place it into the cabinet in order to reduced the introduction of contaminants. The flow of work should proceed across the work surface from clean to contaminated area. 5.2.11 Perform procedure slowly by moving items in and out the cabinet using a straight in and out motion and avoiding side motions. Avoid rapid movements, after placed arms/hands inside the BSC, manipulations should be delayed to permit the cabinet to stabilize and allow the flow of air to remove surface contaminants form your arms/hands. 5.2.12 Don’t block the front grille with paper, equipment etc. As this may cause air to enter the work space area instead of being drawn through the front grille and into the HEPA filter in micro. Arm should be raised slightly and operation should be performed on the work surface at least some inches from the front grille. The middle area is ideal for working. 5.2.13 When work is completed all items within the cabinet should be wiped down the disinfectant and removed from the cabinet, do not use the interior of the BSC as a storage area since stray organisms may become “trapped” and contaminate further experiments. The interior surface of the cabinet should also be cleaned with 70% IPA. Let the blower operate for five minute with no activity inside the cabinet to purge the cabinet of contaminants. 5.2.14 Switch “OFF” the blower and tube light. 5.2.15 Record the activities in Bio-safety usage record as per annexure No. XXX. 5.2.16 Frequency: – every six months by outside party. |
