1.0 OBJECTIVE To lay down a Procedure for Cleaning, Operating and Calibration of Heating Block. 2.0 SCOPE This procedure is applicable for Cleaning, Operation and calibration of heating block in the Quality Control Department of Microbiology Area. 3.0 RESPONSIBILITY • Microbiologist : is responsible for preparation of trends as per SOP(Operation and calibration of heating block). • Manager microbiology : is responsible for ensuring that the SOP is followed correctly. 4.0 ACCOUNTABILITY • Department Head 5.0 PROCEDURE(Operation and calibration of heating block) 5.1 Connect the power cord of the heating block to the electrical main switch. 5.2 Switch ON the mains and red colour switch present on front panel of the heating block. 5.3 Red colour upper side show the present temperature value 5.4 Green colour lower side show the set temperature value. 5.5 If set the temperature press than press upper if temperature increase if temperature decrease presses this button. 5.6 Set the “Heater capacity” Knob at 37°c (±1°c). 5.7 Wait for the temperature to reach the set temperature and stabilize. 5.8 Place the assay tubes into the wells for incubation for the specified time. 5.9 After the incubation, remove the test tubes and switch off the instrument by pressing the Power switch. To read this SOP Click here To read this sop Click here SOP for Operation and qualification of Dry heat sterilizer. 5.10 Cleaning 5.10.1 Clean the external surface of the heating block with lint free wet cloth followed by the 70% IPA before and after working. 5.11 Precaution 5.11.1 Place the heating block on a vibration free surface. 5.11.2 When the incubation start switches off the LAF blower, and during incubation do not disturb the heating block or the table on which it is placed. 5.12 Calibration 5.12.1 Set the required temperature that is 37°c for calibration. 5.12.2 Take 5No’s test tube (10x75mm) and add approximately 200µl of purified water in each tube for performing the calibration. 5.12.3 Place the test tubes in the tube heating block wells as per the diagram and allow to stand for few minutes to stabilize the water temperature within the tubes. 5.12.4 Place a calibrated thermometer on one of the tube for few seconds and record the temperature note down the present value in display temperature of heating block. 5.12.5 Now take out the thermometer, wipe it with a tissue paper to clean the water droplets. 5.12.6 Place the thermometer in another tube for few seconds and record the temperature. Also note down the display temperature if heating block in micro. 5.12.7 Continue this process for all the remaining tubes. 5.12.8 Record all the thermometer reading as well as heating block display temperature in format no. XXX. 5.12.9 Calibration shall be done once in a month ± 05days. 5.12.10 Calibration shall be done by approved external party at annual frequency. 5.13 Discrepancies 5.13.1 In case of any discrepancy affix “Out of order: tag with sign & date and inform engineering department for rectification. 5.13.2 If temperature is found out of range then check, set temperature and if required. |
