1.0 OBJECTIVE: 1.1 To lay down the procedure for cleaning and operation of Microscope in the Microbiology department. 2.0 SCOPE: 2.1 This procedure is applicable for the Microscope available in the Microbiology Lab. 3.0 RESPONSIBILITY: 3.1 Microbiologist personnel is responsible to follow procedure as per the procedure mentioned in this SOP. 3.2 QC Head & microbiology head is responsible for implementation and compliance of the procedure as per this SOP. 4.0 ABBREVIATIONS 4.1 SOP : Standard Operating Procedure 5.0 PROCEDURE: 5.1 CLEANING 5.1.1 Ensure that the microscope is switched Off. 5.1.2 Clean the microscope by using a dry lint free cloth. 5.1.3 Perform the cleaning at least once a day and after each use. To read this SOP Click here SOP for Preparation Of Culture Suspension For Growth Promotion Test. 5.2 OPERATION 5.2.1 Ensure that the microscope and its surrounding area are clean. 5.2.2 Connect the electrical terminals of the microscope into the power socket of the external power supply. 5.2.3 Switch on the microscope by pressing ON/OFF switch to ON position. 5.2.4 Set the required brightness by the brightness control. 5.2.5 Place the specimen prepared on the specimen stage of the microscope in micro lab. 5.2.6 Select the required objective lens by moving the nosepiece. 5.2.7 By turning the adjusting knobs sideways, close the aperture diaphragm approximately half way. 5.2.8 Rotate the coarse adjustment knob until the clear image of the specimen appears. 5.2.9 Rotate the fine adjustment knob for further clarity of the image to appear. 5.2.10 Once the clear image of the specimen appears, place a drop of immersion oil on the specimen and focus the specimen under (100X) objective and follow the steps 5.2.8 and 5.2.9 for the clear image to appear. 5.2.11 After the completion of the work remove the specimen slide and clean the objective lens with tissue paper. 5.2.12 Switch off the microscope by pressing ON/ OFF switch to OFF position and then switch off the external power supply. Note: For wet mount & dry slides, use 5X, 10X and 40X Magnification and for oil immersion use 100X Magnification. |
