1.0 OBJECTIVE To lay down the procedure for operation cleaning and Temperature Monitoring of Refrigerator. 2.0 SCOPE This procedure is applicable for operation cleaning and Temperature Monitoring of Refrigerator in the Quality Control Department of Microbiology Area. 3.0 RESPONSIBILITY • Microbiologist : is responsible for carrying out the established procedure. • Microbiology Manager: is responsible for ensuring that the SOP is followed correctly. 4.0 ACCOUNTABILITY • Department Head To Read this SOP Click here SOP for Maintenance of Dehydrated Media. 5.0 PROCEDURE 5.1 Connect the power plug to the main power sockets and switch on the power supply. 5.2 Set the temperature control knob to on “4” number position. 5.3 In case the temperature (2°c to 8°c) is not achieved, if temperature goes more than 8°c than set the knob 5-6 for coolest if temperature goes less than 2°c than knob set on 1-2. 5.4 Open the door of refrigerator and load the material at desired temperature (2°c to 8°c) and cloth the door. 5.5 Refrigerator have three chamber. 5.6 Limits: – Freezer :- (-2°c to -18°c) Refrigerator :- (2°c to 8°c) Storage bin :- (2°c to 8°c) 5.7 Uses of refrigerator in microbiology laboratory 5.7.1 Laboratory refrigerator can be used for store microbial cultures, slants, culture suspension, water sample, pathogen plates, BET kit and other samples. 5.7.2 Make the entry in format No. XXX on the time of keeping material at refrigerator. 5.8 Cleaning of refrigerator 5.8.1 External cleaning 5.8.1.1 Clean the external surface suing a lint free cloth with 70% v/v IPA. 5.8.2 Internal cleaning 5.8.2.1 Switch off the main refrigerator 5.8.2.2 Remove the plug of the equipment from main socket. 5.8.2.3 Open the door and transfer the material at room temperature. 5.8.2.4 Take out the trays from the equipment at place it in a safe place. 5.8.2.5 With the help of lint free cloth socked with 70% v/v filter IPA trays and material back into the refrigerator 5.8.2.6 Connect the plug of the equipment and place all material in the refrigerator. 5.8.2.7 Switch on the main switch of equipment. 5.8.2.8 Frequency: – fortnightly or if any spillage or required. 5.8.2.9 Microbiologist will ensure that cleaning is proper and record as per format No, XXX. 5.8.2.10 Affix a cleaning status label on instrument. 5.9 Temperature Monitoring. 5.9.1 Refrigerator have three portion, 1) freezer 2) middle chamber and 3) storage bin at the bottom. 5.9.2 Calibrated digital thermometers sensor shall be placed inside the refrigerator chamber and wait about 10min. for the monitoring of the temperature. 5.9.3 After chamber monitoring thermometer sensor placed inside the storage bin and wait about 10min for the monitoring. 5.9.4 After storage bin monitoring digital thermometer sensor placed inside the freezer and wait about 15min. for the monitoring. 5.9.5 Microbiologist’s shall monitor the temperature of chamber, storage and freezer of the refrigerator at frequency of twice in a day. 5.9.6 The noted temperature reading of the refrigerator shall be recorded in the format No. XXX. 5.10 In case if the temperature is not maintained within range after thermoset knob adjusted, then inform to maintenance department for further course of action. 5.11 Temperature mapping shall be done once in a year by external agency. |
