1.0 OBJECTIVE To lay down a Procedure for cleaning, operation and qualification of dry heat sterilizer. 2.0 SCOPE This procedure is applicable for cleaning, operation and qualification of dry heat sterilizer in the Quality Control Department of Microbiology Area. 3.0 RESPONSIBILITY • Microbiologist: is responsible for carrying out the established procedure • Manager microbiology: is responsible for ensuring that the SOP is correctly follow 4.0 ACCOUNTABILITY • Department Head 5.0 PROCEDURE 5.1 Principle: this method is used on objects that are sensitive to moisture. Moisture free heat or dry heat is applied on the surface or objects such that there is denaturation and lysis od protein which leads to oxidative damage, and ultimately the microbial cell dies our or may even burn. To Read this SOP Click Here SOP for Cleaning Operation and Qualification of incubator. 5.2 Cleaning 5.3 Cleaning procedure of incubator. 5.3.1 Daily cleaning: clean the outer surface of incubator with clean and dry lint free cloth. 5.3.2 Fortnightly cleaning. 5.3.1.1 Switch off the main and disconnect the power cord of incubator. 5.3.1.2 Arrange the material between the shelves of same incubator during cleaning. 5.3.1.3 Take lint free cloth and dip purified water contain vessel and clean the incubator trays and inner and outer surface. 5.3.1.4 After cleaning and drying sanitize the inner and outer surface with 70% isopropyl alcohol. 5.3.1.5 After completion of cleaning connect the supply switch on the incubator and keep the material back in the incubator. 5.3.1.6 Fortnightly cleaning details shall be recorded in the format no. XXX. 5.3.1.7 Frequency of fortnight cleaning shall be 15days ± 2days. 5.4 Operation 5.4.1 Switch Insert the plug in the main supply socket and switch it “ON”. 5.4.2 Check that there is any abnormal sound is observed during operation. 5.4.3 Pull up the MCB of DHS in back side of controller. 5.4.4 Pull up the MCB front of DHS one for main and one for fan. 5.4.5 Load the BET test vial for depyrogenation at 200°c for 01hrs. 5.4.6 For glassware, drying set the temperature at 70°c for 01 hrs. 5.4.7 After use off the DHS by down of MCB. |
