1.0 OBJECTIVE To lay down the Procedure for the preparation of filtered 70% IPA for usage in microbiology lab for sanitization. 2.0 SCOPE This procedure is applicable for preparation of filtered 70% IPA for usage in microbiology lab for sanitization in the Quality Control Department of Microbiology Area. 3.0 RESPONSIBILITY • Microbiologist : is responsible for preparation and maintain record as per SOP. • Manager microbiology : is responsible for ensuring that the SOP is followed correctly 4.0 ACCOUNTABILITY • Department Head 5.0 PROCEDURE 5.1 Purpose of filtered IPA 5.1.1 Filtered IPA shall be used in microbiology laboratory as a sanitizing agent for the sanitization of the gloved hands during the microbial testing. 5.1.2 Filtered IPA shall be sprayed on the gloved hands. 5.1.3 The sanitization agent shall be allowed to contact the entire surface of both the gloved hands. 5.1.4 Ensure that the sanitization of the gloved shall be done intermittently during every microbial activity. 5.1.5 The sanitization with filtered IPA shall be done by the microbiologist in the sterility test area and controlled area of the microbiology laboratory. 5.2 Disinfectant preparation. 5.2.1 70% v/v concentration shall be prepared by mixing IPA and water for injection in ratio of 70:30 in a clean beaker at media preparation room. 5.2.2 Preparation details shall be mentioned in format no. XXX. To read this SOP Click here SOP for Environment Monitoring In Microbiology Lab. 5.3 Filtration of 70% IPA. 5.3.1 The all filtration assembly required for the filtration of IPA shall be sterilized in the autoclave at 121-124°c for a period of 30minutes or as per validated cycle. 5.3.2 The prepared 70% v/v IPA solution shall be filtered through 0.22µ membrane inside the microbiological testing room. 5.3.3 In microbiological testing room, collect the sterilized glass filtration assembly and pre-sterilized or sterilized membrane from pass box and keep in under the LAF. 5.3.4 Un-wrap the sterilized outer cover of the all filtration assembly. 5.3.5 Using a blunt tipped incinerated forceps, place sterilized 0.22µ membrane filter on the filter holder. 5.3.6 Connect the filtration assembly to the vacuum source. 5.3.7 Pour the 70% IPA solution to be filtered into the assembled sterilized filtration holder. 5.3.8 Run vacuum pump to filter the 70% IPA solution. 5.3.9 After the entire 70%IPA solution is filtered, stop the vacuum. 5.3.10 Remove the top of the filter holder. 5.3.11 Using a blunt tipped incinerated forceps, remove the membrane filter and discard. 5.3.12 The collected filtered 70% IPA solution in the filtration assembly shall be filled in spray bottles which are used for routine usage. 5.3.13 The spray bottle shall be first rinsed with 70% IPA solution before filling within filtered 70% IPA solution. 5.3.14 The spray bottle filled with filtered 70% IPA solution shall be placed inside the LAF of the controlled area. 5.3.15 All filled spray bottle shall be tagged as per format no. XXX. 5.3.16 The filtered 70% IPA solution shall be used within 48hours or as per hold time study. 5.3.17 In case, if filtered 70% IPA solution is not fully consumed within hold time of it is preparation then unused filtered 70% IPA solution shall be discarded. 5.3.18 The frequency of preparation shall be one day before of hold time of IPA. |
