1.0 OBJECTIVE To lay down a Procedure for cleaning, operation and qualification of incubator. 2.0 SCOPE This procedure is applicable for cleaning, operation and qualification of Incubator in the Quality Control Department of Microbiology Area. 3.0 RESPONSIBILITY • Lab-attendant/Microbiologist: is responsible for carrying out the established procedure • Head of Microbiology : Operation and Qualification of incubator is responsible for ensuring that the SOP is followed correctly. 4.0 ACCOUNTABILITY • Department Head • Head Quality / QA 5.0 PROCEDURE 5.1 Safety considerations: 5.2 The place of installation should be well ventilated. 5.3 Connect proper earthling 5.4 Main plug shall not be touched with wet hands. 5.5 Cleaning procedure of incubator. 5.5.1 Daily cleaning: clean the outer surface of incubator with clean and dry lint free cloth. 5.5.2 Fortnightly cleaning. 5.5.2.1 Switch off the main and disconnect the power cord of incubator. 5.5.2.2 Arrange the material between the shelves of same incubator during cleaning. 5.5.2.3 Take lint free cloth and dip purified water contain vessel and clean the incubator trays and inner and outer surface. 5.5.2.4 After cleaning and drying sanitize the inner and outer surface with 70% isopropyl alcohol. 5.5.2.5 After completion of cleaning connect the supply switch on the incubator and keep the material back in the incubator. 5.5.2.6 Fortnightly cleaning details shall be recorded in the format no. XXX “Incubator Cleaning Record”. 5.5.2.7 Frequency of fortnight cleaning shall be 15days ± 2days. 5.5.2.8 Contingency plan shall be followed to mitigate the risk of incubator failure and incubation of sample if nay incubator failure occurred, the samples under incubation shall be transferred in other incubator (operated at same temperature). To Read this SOP click here OP for Entry and Exit in Microbiology Testing Area. 5.6 Operation and monitoring of the temper true in incubator 5.6.1 Inset the plug in the main supply socket and switch it on. 5.6.2 Switch on the mains of the incubator to “ON” position; the main indicator will show red light. 5.6.3 Switch “ON” the fan switch for air circulation. 5.6.4 Press UP- Arrow () or down arrow () keys to set desired valve of set point in green display. Press mode for one second to storing new set value. If this key is not pressed, then old valve will appear again. Set the temperature of the incubator with the help of digital temperature controlled keys symbol as ( and ). Incubator No. Set condition XXX 42-44°c CCC (as per requirement) Note: Incubator No. XXX validate on different temperature (55-60°C, 42-44°c and 30-35°c). This incubator used for incubation of biological indicator and use as standby. 5.6.5 Allow the incubator to stabilize for 30minutes; check the desired temperature during this period. 5.6.6 The safety temperature controlled is set to higher temperature if the required incubation temperature is more than the ambient and set it to lower temperature if the required incubation temperature is less than the ambient. 5.6.7 The voltage indicator shows the voltage supplied to the incubator, if it is high or low then incubator will be switched off automatically. 5.6.8 When the incubator is not required first switch off the digital controlled alarm button and then the main switch to the “OFF” position, then stop the main supply. 5.6.9 Pull the door of incubator and keep the desired material for the period required as per the test conditions. 5.6.10 After keeping material, close the door. 5.6.11 Temperature of shall be monitored twice in shift and by observing the temperature displaying on the display of incubator and observation shall be recorded in format No. XXX. 5.6.12 The safety temperature controlled is set to higher temperature, if the temperature required for the incubation is more than the ambient and set it to lower temperature if the required for the incubation is less than the ambient temperature. 5.6.13 The voltage indicator shows the voltage supplied to the incubator, if it is more or less, than incubator will be switched off automatically. 5.6.14 When the incubator is not required first switch off the incubator MCB and then main switch to the main OFF position, then stop the main supply. 5.6.15 In case of any problem with the working of the incubator, inform the maintenance department. |
